Its construction took place at the beginning of the 17th century, its founder being Pedro de Toledo y Colonna, V Marquis of Villafranca, II Duke of Fernandina and Prince of Montalbán. The foundation is due to the wishes of his daughter María to be a nun. It is built in the place where the pilgrim hospital of Santiago was located. The church has a single nave, with a lunette vault and a blind dome. Its altarpiece is made up of three reliefs from the 17th century that represent The Annunciation, the Birth of Christ and the Betrothal of Saint Catherine. In the center of the altarpiece there is a magnificent large monstrance brought from Italy.

The church, along with other rooms of the cloister, houses a collection of Flemish paintings from the year 1601. The collection is known as the hermits of the Annunciation, since each painting has a hermit as the protagonist. Furthermore, inside the church is kept the body of San Lorenzo de Brindis, Doctor of the Church, declared as such by Pope John XXIII in 1959. At the foot of the church is the Pantheon of the marquises of the town, highlighting the funeral mound of the V Marquis and his daughter María.
